Use the caulking gun to apply sealant on the inner lip of the gutter segment known as the downhill.
Apply additional sealant inside the gutter at the seam pressing the sealant into the entire crevice.

If there is a gap in the joint you should use your cardboard or plastic spreader to force sealer into the joint.
Replace all pieces that make the seam and then finish by re adding the screws.
